A convenient synthetic strategy toward N-acylguanidines via a sequential one-pot multicomponent carbonylation/amination reaction has been developed. The compounds were readily obtained via an N-cyanobenzamide intermediate formed from the Pd(0)-catalyzed carbonylative coupling of cyanamide and aryl iodides or bromides. Subsequent amination with a large variety of amines provided the final N-acylguanidines, with the overall formation of one C-C and two C-N bonds, in moderate to excellent yields. The substrate scope was found to be wide and the methodology was used to produce over 50 compounds, including 29 novel molecules. Furthermore, three separate nitrogen-containing heterocycles were prepared from the N-acylguanidines synthesized using the developed multicomponent, carbonylative method.

The recently introduced pump-probe fragmentation action spectroscopy reveals a unique observation of excited state vibrational coherence (430-460 fs) in the isolated metal complex [Ag2(Cl)(dcpm)2)]+ (dcpm = bis(dicyclohexylphosphino)methane) containing the [Ag2Cl]+ scaffold. After photoexcitation by an 1XMCT transition (260 nm) in an ion trap, an unexpected correlation between specific fragment ions (loss of HCl/Cl- vs loss of dcpm) and the phase of the wave packet is probed (1150 nm). Based on ab initio calculations, we assign the primary electronically excited state and ascribe the observed coherences (72-78 cm-1) to contain predominantly Ag-Ag stretch character. We propose specific probe absorption and vibronic coupling at the classical turning points to switch remarkably early on between the different fragmentation pathways. The overall excited state dynamics are fitted to a multiexponential decay with time constants: 0.2-0.4/3-4/19-26/104-161 ps. These findings open new perspectives for further dynamics investigations and possible applications in photocatalysis.

Pregnane X receptor (PXR) is a ligand-dependent transcription factor that is considered to be a potential therapeutic target for multiple diseases. Herein, we report the development and structure-activity relationship studies of a new series of hPXR agonists. Focusing on our recently developed silanol-sulfonamide scaffold, we developed the potent hPXR agonist 28, which shows good selectivity over hLXRalpha and beta, hFXR, and hRORalpha and gamma. Examination of the structure-activity relationship suggested a possible strategy to manipulate the selectivity. Docking simulation indicated the presence of an additional binding cavity and polar contacts in the ligand-binding pocket of hPXR. This information should be helpful for the future development of more potent and selective hPXR ligands.

A series of silica-tethered Ru complexes were easily prepared and further analyzed by sophisticated analytical techniques such as FTIR, N2physisorption, ICP-OES and XPS analysis. Nowadays, computational studies are very important for the elucidation of reaction mechanisms and selectivity of complex reactions. However, traditional computational methods usually require an estimated reaction path, mainly driven by limited experimental implications, intuition, and assumptions of stationary points. However, the artificial force induced reaction (AFIR) method in the global reaction route mapping (GRRM) strategy can be used for unbiased and automatic reaction path searches for complex reactions. In this account, we highlight applications of the AFIR method to a variety of reactions (organic, organometallic, enzymatic, and photochemical) of complex molecular systems. In addition, the AFIR method has been successfully used to rationalise the origin of stereo- and regioselectivity. The AFIR method can be applied from small to large molecular systems, and will be a very useful tool for the study of complex molecular problems in many areas of chemistry, biology, and material sciences.
