Antitumor activity of bis(diphenylphosphino)alkanes, their gold(i) coordination complexes, and related compounds

Bisphosphines related to bis(diphenylphosphino)ethane (dppe) and their gold complexes are described that are active in a spectrum of transplantable tumor models. When administered ip on days 1-5 at its maximally tolerated dose (MTD) of 40 mumol/kg, dppe reproducibly gives 100% increase in life span (ILS) in mice bearing ip P388 leukemia. Coordination of chlorogold(I) to each phosphine in dppe gave a complex that had similar activity but at a much lower dose level than dppe; the MTD for the gold(I) complex was 7 mumol/kg. Among other metal complexes of dppe, the Au(III) complex was active (>50% ILS) whereas Ag(I), Ni(II), Pt(II), Pd(II), and Rh(I) complexes were inactive. Among dppe analogues, replacement of phenyl groups with ethyl or benzyl groups resulted in inactivity for both ligands and the corresponding gold complexes whereas substitution with cyclohexyl or heterocyclic ring systems yielded ligands and/or gold complexes with antitumor activity. Among substituted-phenyl dppe and dppe(AuCl)2 analogues, 3-fluoro, 4-fluoro, perdeuterio, 4-methylthio, and 2-methylthio analogues were active; 4-methyl, 3-methyl, 4-methoxy, 4-dimethylamino, and 4-trifluoromethyl analogues were marginal or inactive. Analogues in which the ethane bridge of dppe or dppe(AuCl)2 was varied between one and six carbons, unsaturated or substituted, revealed that activity was maximal with ethane or cis-ethylene. Compounds with good P388 activity were also active in other animal tumor models.

Aryl-CF3 Coupling from Phosphinoferrocene-Ligated Palladium(II) Complexes

This article describes a detailed investigation of ligand effects on Ph-CF3 coupling from phosphinoferrocene-ligated PdII(Ph)(CF3) complexes. This study reveals that increasing the size of the phosphine substituents results in an enhanced rate of Ph-CF3 coupling, with (DtBPF)Pd(Ph)(CF3) (DtBPF = 1,1?-bis(di-tert-butylphosphino)ferrocene) being the most reactive complex. The mechanism of Ph-CF3 bond formation from both (DtBPF)Pd(Ph)(CF3) and (DiPrPF)Pd(Ph)(CF3) (DiPrPF = 1,1?-bis(diisopropylphosphino)ferrocene) was interrogated experimentally and computationally. These studies implicate a pathway involving concerted Ph-CF3 bond-forming reductive elimination from the four-coordinate PdII centers. An alternative pathway involving alpha-fluoride elimination and subsequent PhF2C-F coupling from PdII(CF2Ph)(F) intermediates was also evaluated but was ruled out based on DFT as well as the independent synthesis and reactivity studies of (DiPrPF)Pd(CF2Ph)(F).

Reductive Elimination of C6F5-C6F5 from Pd(II) Complexes: Influence of alpha-Dicationic Chelating Phosphines

We report the synthesis and characterization through NMR and X-ray techniques of a series of [Pd(C6F5)2(PP?)] complexes constituted by diphosphine chelating ligands of different nature and evaluate the rates for the challenging reductive elimination of C6F5-C6F5. By virtue of their very weak donor properties, dicationic ancillary ligands effectively promote the desired transformation. Density functional theory (DFT) calculations were performed to rationalize these findings. The Pd(0)-complexes formed after the elimination step could not be isolated because the Pd(0) center has a tremendous tendency to insert into one of the P-C+ bonds of the alpha-cationic ligands rendering Pd(II)-phosphinidene complexes. The same behavior was observed for Ni(0) species.

Palladium-Catalyzed Synthesis of alpha-Carbonyl-alpha?-(hetero)aryl Sulfoxonium Ylides: Scope and Insight into the Mechanism

Despite recent advances, a general method for the synthesis of alpha-carbonyl-alpha?-(hetero)aryl sulfoxonium ylides is needed to benefit more greatly from the potential safety advantages offered by these compounds over the parent diazo compounds. Herein, we report the palladium-catalyzed cross-coupling of aryl bromides and triflates with alpha-carbonyl sulfoxonium ylides. We also report the use of this method for the modification of an active pharmaceutical ingredient and for the synthesis of a key precursor of antagonists of the neurokinin-1 receptor. In addition, the mechanism of the reaction was inferred from several observations. Thus, the oxidative addition complex [(XPhos)PhPdBr] and its dimer were observed by 31P{1H} NMR, and these complexes were shown to be catalytically and kinetically competent. Moreover, a complex resulting from the transmetalation of [(XPhos)ArPdBr] (Ar = p-CF3-C6H4) with a model sulfoxonium ylide was observed by mass spectrometry. Finally, the partial rate law suggests that the transmetalation and the subsequent deprotonation are rate-determining in the catalytic cycle.

Treatment of the chloro-bridged dinuclear compound [{Pd[4-(COH)C6H3C(H)=N(Cy)-C2,N]}(mu-Cl)]2 (1) with tertiary diphosphines in 1:1 molar ratio gave [{Pd[4-(COH)C6H3C(H)=N(Cy)-C2,N] (Cl)}2(mu-Ph2PRPPh2)] (R: CH2, 2; CH2CH2, 3; (CH2)4, 4; (CH2)6, 5: Fe(C5H4)2, 6; trans-CH=CH, 7; C?C, 8) with the diphosphine in a bridging mode. When the reaction was carried out in a 1:2 molar ratio in the presence of NH4PF6, the compounds [Pd{4-(COH)C6H3C(H)=NCy-C2,N} (Ph2PRPPh2-P,P)][PF6] (R: CH2, 9; CH2CH2, 10; (CH2)4, 11; (CH2)6, 12; Fe(C5H4)2, 13; 1,2-C6H4, 14; cis-CH=CH, 15; NH, 16) with the diphosphine chelated to the palladium atom, were obtained. 1H, 31P-{1H} and 13C- {1H} NMR, IR and mass spectroscopic data are given. The crystal structure of compound 2 has been determined by X-ray crystallography.
