We present an expedient and economical route to a new spiroketal-based C-2-symmetric chiral scaffold, termed SPIROL. Based on this spirocyclic scaffold, several chiral ligands were generated. These ligands were successfully employed in an array of stereoselective transformations, including in iridium-catalyzed hydroarylations (up to 95% ee), palladium-catalyzed allylic alkylations (up to 97% ee), intermolecular palladium-catalyzed Heck couplings (up to 94% ee), and rhodium-catalyzed dehydroalanine hydrogenation (up to 93% ee).

Optically active helical polyisocyanides bearing chiral phosphine pendants: Facile synthesis and application in enantioselective Rauhut-Currier reaction

Three novel enantiopure phenyl isocyanide monomers with BH3-protected phosphine functional group were designed and synthesized. Polymerization of these monomers using a alkyne-Pd(II) complex as a catalyst led to the formation of respective helical polyisocyanides in high yields with controlled molecular weights (M (n)s) and narrow molecular weight distributions (M (w)/M (n)s). Removing the protecting BH3 groups afforded helical poly(phenyl isocyanide)s bearing phosphine pendants. Thanks to the chiral induction of monomer, the isolated helical polyisocyanides showed high optical activity, as revealed by circular dichroism (CD) and absorption spectroscopies and polarimetry. The helical structures of these polymers were quite stable in various organic solvents with different polarities and in a wide temperature range. Moreover, these helical polymers could be used as organocatalysts and showed good performance in enantioselective cross Rauhut-Currier reaction. The enantiomeric excess (ee) values of the isolated products of cross Rauhut-Currier reaction could be up to 90%. The polymer organocatalysts could be easily recovered from the reaction mixtures and reused at least five times in the reaction without significant loss of their enantioselectivities and catalytic activities.

Herein, we report the first catalytic and asymmetric Staudinger-aza-Wittig reaction for the desymmetrization of ketones. This highly enantioselective transformation proceeds at room temperature to provide high yields-even on multigram scales-of nitrogen heterocycles featuring a chiral quaternary center. The products of this reaction are potential precursors for the synthesis of pharmaceuticals. A commercially available small P-chiral phosphine catalyst, HypPhos, induces the asymmetry and is recycled through in situ reduction of its oxide, mediated by phenylsilane in the presence of a carboxylic acid. The efficiency, selectivity, scalability, mild reaction conditions, and broad substrate scope portend that this process will expedite the syntheses of chiral heterocyclic amines of significance to chemistry, biology, and medicine.

Development of a Scalable, Chromatography-Free Synthesis of t-Bu-SMS-Phos and Application to the Synthesis of an Important Chiral CF3-Alcohol Derivative with High Enantioselectivity Using Rh-Catalyzed Asymmetric Hydrogenation

A chromatography-free, asymmetric synthesis of the C2-symmetric P-chiral diphosphine t-Bu-SMS-Phos was developed using a chiral auxiliary-based approach in five steps from the chiral auxiliary in 36% overall yield. An air-stable crystalline form of the final ligand was identified to enable isolation of the final ligand by crystallization to avoid chromatography. The current approach relies on the use of chiral bifunctional phosphine-carboxylate ligands and delivers diverse polycyclic compounds in high yield and with good to excellent enantioselectivity. The chiroptical properties of the obtained products were investigated, and some of them were found to have a strong ellipticity and an emission band located in the visible region.

This study describes the development of new chiral bifunctional ligands based on a binaphthyl scaffold which incorporates both a phosphine and a carboxylic acid moiety. The optimal ligand provided high yields and enantioselectivities for a desymmetrizing C(sp(2))-H arylation leading to 5,6-dihydrophenanthridines, whereas the corresponding monofunctional ligands showed low enantioselectivities. The bifunctional system proved applicable to a range of substituted dihydrophenanthridines, and allowed the parallel kinetic resolution of racemic substrates.
