Disclosed is an improved treatment for men with benign prostatic hyperplasia (BPH), involving combination therapy of a 5alpha-reductase inhibitor, e.g. a 17beta-substituted 4-azasteroid, a 17beta-substituted non-azasteroid, 17beta-acyl-3-carboxy-androst-3,5-diene, benzoylaminophenoxybutanoic acid derivative, fused benz(thio)amide or cinnamoylamide derivative, aromatic 1,2-diethers or thioethers, aromatic ortho acylaminophenoxy alkanoic acids, ortho thioalkylacylaminophenoxy alkanoic acids, pharmaceutically acceptable salts and esters thereof, and particularly finasteride, in combination with an alpha1 -adrenergic receptor blocker, i.e., terazosin. The combination provides therapy at the molecular level for the underlying cause of the disease as well as providing symptomatic relief. Pharmaceutical compositions useful for treatment are also disclosed.

Facile photoreductive protocols have been developed to remove benzyl O-protective groups from oxyarene N-heterocycles at positions capable for 2-/4-O-pyridine-2-/4-pyridone tautomerism. Blue light irradiation, a [Ru] or [Ir] photocatalyst, and ascorbic acid in a water-acetonitrile solution debenzylates a variety of aryl N-heterocycles cleanly and selectively. Ascorbic acid has two functions in the reaction. On the one hand, it protonates the N-heterocycles that reduces their reduction potentials notably and on the other hand it acts as a sacrificial reductant. Reduction potentials and free energy barriers calculated at the CPCM-B3LYP/6-31+G? level can predict the reactivities of the studied substrates.

Structural study of four benzyloxybenzoic acids and benzyloxyanilines using X-ray powder diffraction: Interplay of strong and weak intermolecular interactions

Crystal structures of two benzyloxybenzoic acids (1 and 2) and two benzyloxyanilines (3 and 4) have been solved from laboratory X-ray powder diffraction data using the direct space methodology. The refinements of structures 1, 2, 3 and 4 have been carried out following the Rietveld method. The nature of intermolecular interactions in 1-4 has been analyzed through the Hirshfeld surfaces and 2D fingerprint plots. The COOH functional group generates a cyclic R22(8) synthon via intermolecular O-H…O hydrogen bonds in 1 and 2. An interplay of O-H…O and C-H…O hydrogen bonds, and C-H…pi interactions in 1 and 2 facilitates the formation of three-dimensional supramolecular frameworks in 1 and a one-dimensional step-like architecture in 2. While the combination of N-H…pi and C-H…pi interactions link the molecules in 3 into parallel columns along the [100] direction, no such molecular assembly has been observed in 4. Hirshfeld surface analyses of 1-4 as well as a few related benzyloxybenzoic acid and benzyloxyaniline derivatives retrieved from the Cambridge structural database (CSD) indicate that about 90% of Hirshfeld surface areas in this class of compounds are due to H…H, C…H and O…H contacts.

Synthesis and structure-activity relationships of N-(4-amino-2,6-diisopropylphenyl)-N’-(1,4-diarylpiperidine-4-yl)methylureas as anti-hyperlipidemic agents

Based on 1,4-diarylpiperidine-4-methylureas, a new class of ACAT inhibitors, we examined in the study the SAR of a series of compounds prepared by replacing the substituent at the three aromatic parts. Introduction of long alkoxy group onto the phenyl moiety at the B-part was effective in improving both the inhibitory activity for ACAT and the up-regulatory activity for LDL-R expression. Particularly, 3-hydroxypropoxy group (43) on the phenyl moiety of B-part led to improved solubility, while keeping both biological activities. Compound 43 inhibited ACAT activity with an IC50 value of 18 nM, which is superior to that of a known ACAT inhibitor, CI-1011. In addition, compound 43 revealed an LDL-R up-regulatory activity comparable to that of SMP-797. We therefore expect this compound to be a novel ACAT inhibitor.

Design, synthesis, and evaluation of N-phenyl-4-(2-phenylsulfonamido)-benzamides as microtubule-targeting agents in drug-resistant cancer cells, displaying HDAC inhibitory response

Microtubule-targeting agents (MTA) have enjoyed significant clinical success for decades. However, several mechanisms may cause inactivation of such drugs, leading to acquired resistance in patients treated with them. Therefore, drugs containing a stilbene-like skeleton and possessing dual inhibitory activity may provide a new and differentiated treatment for patients to overcome challenging acquired resistance. A new compound (16c) displays promising anticancer activity with GI50 of 22 ¡À 2 and 12 ¡À 0.1 nM in vincristine-resistant nasopharyngeal (KB-Vin) cancer cells and etoposide-resistant nasopharyngeal (KB-7D) cancer cells and is better than vincristine, etoposide, ABT-751, and MS-275. A mechanistic study revealed that 16c interferes with the cell cycle distribution and induces cell cycle arrest at the G2/M phase and severe mitotic spindle defects followed by apoptosis. In addition, it produces much more significant cytotoxicity than vincristine and etoposide in the corresponding resistant cells, indicating that it may be a promising candidate to overcome drug resistance in cancer cells. Compound 16c also displays inhibitory activity against HDAC 1 and HDAC 2 with IC50 values of 1.07 muM, and 1.47 muM, respectively. These findings may lead to a new type of structural motif for future development of drugs that could overcome acquired resistance to MTAs.

Novel 5-hydroxytryptamine (5-HT3) receptor antagonists. II. Synthesis and structure-activity relationships of 4,5,6,7-tetrahydro-1H-benzimidazole derivatives

A novel series of 4,5,6,7-tetrahydro-1H-benzimidazole derivatives 4, 5, 6 and 7 was prepared and evaluated for activities as 5-hydroxytryptamine (5- HT3) receptor antagonists which may be useful for the treatment of irritable bowel syndrome (IBS) as well as nausea and vomiting associated with cancer chemotherapy. These compounds were designed by modifying the aromatic- carbonyl part of N-(2-methoxyphenyl)-4,5,6,7-tetrahydro-1H-5- benzimidazolylcarboxamide 3, leaving the imidazole moiety unchanged as the amine part. The indole derivatives 7d, g, h and indolizine derivatives 7k, 1 were found to be highly potent on the von Bezold-Jarisch (B.J.) reflex test with ID50 values of below 0.1 mug/kg, and the indoline derivative 6c, indole derivatives 7a, d, g, benzofurane derivative 7j and indolizine derivative 7k were observed to be very potent on the colonic contraction with IC50 values of below 0.1 muM. In particular, 71 was the most potent on the B.J. reflex (ID50 = 0.018 mug/kg), approximately 200 and 50 times more potent than ondansetron 1 and granisetron 2, and 7k was the most potent on the colonic contraction (IC50 = 0.011 muM), approximately 70 and 6 times more potent than 1 and 2, respectively.
