N-(2-Methoxyphenyl)-5-methyl-2-(5-morpholin-4-yl-3,4′-bipyridin-2′-yl)-1H-imidazol-4-amine trifluoroacetate salt To a degassed mixture of 2′-(4-iodo-5-methyl-1H-imidazol-2-yl)-5-morpholin-4-yl-3,4′-bipyridine (30 mg, 0.067 mmol, from Example 257, Step 2), di-tert-butyl(2′,4′,6′-triisopropyl-3,6-dimethoxybiphenyl-2-yl)phosphine (2.0 mg, 0.0040 mmol), tBuBrettPhos Pd G3 (3.4 mg, 0.0040 mmol), and 2-methoxyaniline (9.7 muL, 0.080 mmol) in THF (0.25 mL) was added 1.0 M LHMDS (lithium bis(trimethylsilyl)amide) in THF (150 muL, 0.15 mmol). The mixture was sealed and heated at 70 C. for 2 hours. Upon cooling to room temperature, the reaction mixture was quenched by the addition of 1N HCl (1 mL). The mixture was diluted with ACN/MeOH, filtered, and purified via preparative HPLC/MS (pH 2). Yield: 8.5 mg. 1H NMR (400 MHz, d6-DMSO) delta 8.88 (d, J=5.2 Hz, 1H), 8.58-8.52 (m, 2H), 8.50 (d, J=2.2 Hz, 1H), 8.10-8.02 (m, 1H), 7.87-7.78 (m, 1H), 7.45 (br s, 1H), 7.05-6.95 (m, 1H), 6.86-6.73 (m, 2H), 6.64-6.53 (m, 1H), 3.89 (s, 3H), 3.85-3.72 (m, 4H), 3.43-3.27 (m, 4H), 2.21 (s, 3H). LCMS(M+H)+: 443.1.
